In canon, the only interest that Sam has shown attraction to are women. Here they are in somewhat canonical order:
Jess - 1.01 "Pilot". Romantic/Sexual - they were together for at least 18 months [He tells Dean in 1.13 "Route 666" that he had lied to Jessica for ‘a year and a half’ about what their family did] and was going to propose to soon per the yellow Eye Demon who was possessing John at the time [John (turns back to Dean): You know, I never told you this, but Sam was going to ask her to marry him. (he backs up toward Sam) Been shopping for rings and everything. ]. Sam has nightmares about her death for weeks after (and before) her death, and her death is why he turns back to hunting with his brother. She also appears as Sam's fiancee in Dean's dream world in 2.19 "What is and What Never Should Be", and Lucifer takes on her face to try and gain Sam's trust in 5.03 "Free to be You and Me".
Sarah - 1.19 "Provenence". Date/Kiss. Dean was trying to get his brother to hook up but it never went that far. She is mentioned in the following episode for a moment and never mentioned again.
Meg - 1.11 "Scarecrow" - 1.16 "Shadow". Friendship/Crush. That is until Sam learns she's a demon trying to kill him, Dean and John. Then disgust/loathing.
Madison - 2.17 "Heart" - Romantic/Sexual. first woman Sam has had a fully romantic interest in since Jess' death. She is also discovered to be the werewolf they are hunting, and she asks Sam to kill her at the end of the episode which he does because he loves her and doesn't want her to suffer that life. She's also mentioned at various times over the next seasons (4.14 "Sex and Violence, 4.17 "It's a Terrible Life", etc.
Ruby - Seasons 3 & 4 - Romantic/Sexual. The relationship part of this duo starts during the season break (not revealed until 4.09 "I Know What You Did Last Summer") and Sam also becomes addicted to her blood and her in the process. However, it was all done to manipulate Sam into killing Lilith and freeing Lucifer and Sam holds her while Dean kills her in 4.22 "Lucifer Rising".
Bela - 3.10 "Dream a Little Dream of Me" - Fantasy Sexual/Crush. Bela uses dreamroot to get into Sam's head while he is asleep and they end up having sex until Dean wakes him up prematurely. He is then flustered when she appears minutes later.
Cara - 4.14 "Sex and Violence" - Sexual. She hits on him, he flirts right back, wham bam thank you ma'am right in her office and he doesn't even call the next day.
Lindsey - 5.03 "Free to Be You and Me" - Friendship. When Sam gets a job at the bar, he's trying to keep his head down low but she wants him to open up and is infatuated with him. When the hunters come back they hold her hostage knowing that Sam won't let her get hurt. There was supposed to be a sex scene between them in the original script, but was cut before the aired episode.
There are also examples in canon that support Sam not being homosexual, nor wanting a sexual relationship with his brother:
Against being gay:

Larry: Let me just say - we accept homeowners of any race, religion, color, or sexual orientation. [SAM and DEAN realize what he is trying to say.]
Dean: We're brothers. [LARRY seems slightly embarrassed.]
2.11 "Playthings "
Dean: Of course, the most troubling question is why do these people assume we're gay?
Sam: Well, you are kinda butch. Probably think you're overcompensating.
2.19 "Folsom Prison Blues"
Dean: (in a low voice to SAM, who is standing behind him) My roommate doesn't say much, how's yours?
Sam: (in a low voice) Just keeps staring at me... In a way that makes me really uneasy.
Dean: It sound like you're making new friends.
Against sleeping with his brother:
4.18 "Monster at the End of this Book". While the dialogue is mainly on Dean's end, Sam's voice and body language show he is in agreement.
Dean: There's Sam Girls and Dean Girls and...What's a slash fan?
Sam: As in Sam slash Dean together.
Dean: Like together, together? They do know we are brothers right?
Sam: Doesn't seem to matter.
Dean: Well that's just sick!
He also has shown that he is uncomfortable with women over a certain age, as he was once paired up with a 70+ year old woman, Gertie, for a cover operation in3.06 "Red Sky at Morning" and expressed his displeasure for the entire time while Gertie made very openly sexual advances at him.
Dean has also accused Sam of having a thing for monsters in 4.14 "Sex and Violence" - Madison (werewolf), Ruby (demon), and at that point believing Cara was a siren - which upsets Sam.
» Personality: Sam has gone through a lot in the last five years, and it's had an effect on his personality. But the core elements of Sam have never changed, even if they have altered a bit.
Sam is intelligent. The kid got a 174 on his LSAT! It's always been shown that Sam was "geekboy" and did most of the research in the pairing, but beyond that he is also the voice of reason at times when Dean is getting caught up in the moment. He's the one who can tell when they need to take a step back and look at something again. However, he does have a blind spot when someone who he cares about is in danger, and he will throw caution to the wind and go after something without thinking. It's in those moments where Sam tends to make some of his greater mistakes.
Sam is a lover. When it comes to women, Sam isn't a love them and leave them type (with Cara as an exception). He has a sweet smile, and knows how to make any woman feel at ease - whether he's trying to get information from them for a case, or soothing a victim from an attack. Dean has even deliberately had Sam use his "puppy dog thing" on cases. When it comes to actual relationships, Sam has been shown that he needs to form some kind of relationship with a woman before he is able to open up with his feelings to them. He can even be shy and sometimes clueless that he is being flirted with. When a girl does get past his walls, he is a passionate man who will defend his woman from anything that could harm them - even against his own brother like in the case with Ruby.
Sam is an addict. Not just in the sense of his addiction to the power that he gets from the demon blood, but Sam has an addictive personality. He has a dangerous co-dependency to his brother. Without having the constant companionship of his brother, Sam is unable to function. In 3.11 "Mystery Spot", Dean is killed and we are shown that Sam becomes desensitized to the world (compared to the characters in Full Metal Jacket) for the four months until the Trickster fixes the time-line. When Dean really dies at the end of season 3, and it's revealed what happens to him in 4.09 "I Know What You Did Last Summer" that he becomes unhinged, suicidal, and an alcoholic until Ruby steps in to become his new "drug" with sex and attention, a direction to move, and eventually the blood and power which keeps him there even after Dean comes back.
Sam is a freak. He is human with demon blood. He's been told that Dean might have to kill him. The angels consider him an abomination. He's been told that he is special. He is the chosen one. He is Lucifer's vessel. As the years have gone by since Sam lost his dream of "being normal" with Jess, he has just faced more and more facts that he was different. He never wanted to be a hunter, and yet that is what he is in his blood. He has powers that he hates to use, but he will to try and save people. He's also working on coming to terms with the realization that being a freak doesn't mean that he is damned or wrong.
Sam is a hero. Everything Sam has done as a hero is to help save people. Whether it's trying not to let his brother die, or saving the nameless man on the street, Sam hates seeing people suffer. Part of this comes from being a victim himself from losing the love of his life - Jess - because of who he is. Some of it comes from feeling that he needs to repent for the demon blood in his veins and to change his destiny ("No, Dean, you don't understand, all right? The more people I save, the more I can change!"- 2.11 "Playthings"). But Sam also is just a naturally sweet guy who wants to do the right thing. However, Sam also learns the hard way that the road to hell really is paved with good intentions. And he isn't afraid to make the ultimate sacrifice of himself to save the world, as long as he knows that his brother will get a chance to have a better life for it.
To sum up, Bobby said it best in 5.21 "Two Minutes to Midnight":
Bobby: Back at Niveus, I watched that kid pull one civilian out after another, he must've saved ten people. Never stopped, never slowed down. We're hard on him, Dean. We've always been. But, in the meantime, he's been running into burning buildings since he was - what? Twelve?
Dean: Pretty much...
Bobby: Look, Sam's got a... darkness in him, I'm not saying he don't. But he's got a helluva lot of good in him too.
Dean: I know.
Bobby: Then you know Sam will beat the Devil or die trying. That's the best we can ask for.
» Appearance: Sam is 6’4” barefoot and is often described by his brother as a Sasquatch. In the last two years he has started to grow into his body, losing the beanpole look to have his shoulders broaden as well as definite muscle development in his arms, back, shoulders and chest. When he smiles he has deep dimples in his cheeks. He has hazel “soulful” eyes and wears his brown hair long so that it tends to fall in front of his eyes a lot of the time. He tends to wear blue jeans, a button down shirt with a tee-shirt underneath, a jacket or hoodie, and work boots. He wears no significant jewelry, but has a tattoo over his left pec that is a demon anti-possession ward which he and Dean both got sometime after Sam was possessed by Meg in 2.14 "Born Under a Bad Sign".

[the device is set up, and you can see Sam walking away from it to sit on the edge of the bed. Like any college boy, he remembers how to take a sheet and make it into a toga, and the red silk goes well with the black collar with red etchings in it. He runs a hand through his hair, trying not to look distressed and worried about his situation]
Alright, I have no idea what's going on, but if someone is out there, I could use some help. Or information. [he looks down at his toga] Or at least know where my clothes might be.
[he reaches to turn it off, then stops a moment] And Dean, I don't know if you can hear this, or if you're even... wherever this place is... but if you can, tell me how to find you. I... yeah. [he nods to himself, biting his lower lip nervously before turning the recording function off so he could figure out how to post it next]

They say life flashes before your eyes when you're dying. Sam, however, had already died a few times in his short life and knew it was a bunch of crap. All you got was blackness, like a large pit opening up under your feet and you jump in and the light fades away as you fall down... down... down...
But once you're dead, then you get to remember the good times. Your memories get to play out in front of your eyes for eternity as a piece of heaven or hell that you lived. And seeing that Sam was trapped in a box for eternity while Lucifer and Michael gave each other the silent treatment, Sam had no problem just letting his consciousness slip away to remember a good time. The good times; the time he got for such a short instance to believe that he was normal.
Sam...
Five years. It had been five years since he had been with Jessica and yet he could remember every inch of her body like a map burned into his soul. How their bodies had fit together so perfectly. The feel of her breath on his neck, whispering his name as they moved together. He remembered how her fingers would grip his hair - she liked it long and he wasn't complaining - just as another orgasm would start to show it's head. He always made sure she came first so that they didn't have to stop right away. Just lay there, inside of her waiting until she gave that little smile and they would roll over so she was on top and they would take their time until Jess was ready again and Sam wouldn't hold back then. They held hands when they came sometimes to just keep that bond between them, and then she would stretch out on his chest and lay there as their heartbeats pounded together at the same pace.
It was that heartbeat that woke Sam up from his beautiful memory. It was slow, steady, and comforting as much as it was wrong. He was dead and shouldn't have one. Nor should he be breathing and as that recognition hit his brain he opened his eyes and sat up in the... extremely lavish hotel room? He looked around in confusion, rubbing at his eyes because he still felt fuzzy and tired. His hips were sore as he shifted, and just a bit sticky.
His eyes roamed around the room, a fleeting moment of perhaps having Jess there with him as well, but he was alone. And naked. Way too naked for being in a strange place when he was supposed to be in Lucifer's cage in hell. Dean had painted Sam a good picture of hell and this? This wasn't anything like that.
He saw the open window and carefully slid out of bed, pulling the sheet with him to wrap around his waist to at least preserve some kind of modesty. Walking over, he looked out to see a beautiful beach with gentle waves lapping at the shore - a picture of peace and serenity that Sam's only seen on television. It doesn't help with the confusion, though, so he continues to look around. He starts to check in drawers and under the bed, trying to find something that will tell him where he is, or more importantly, where his clothes are.
As he stood up in front of the dresser, he managed a look at his reflection in the mirror and then leaned forward as he reached up to touch the black collar around his neck. He tilted his neck up and tried to read the etchings that were inscribed in a blood red color, but he couldn't recognize any of them. His fingers tried to find a clasp to take it off with, but he couldn't find any way to remove the thing.
Sighing, Sam turned and leaned against the dresser and just looked around again in disbelief. "Where the hell am I?"
